- Our initial sequencing strategy is to sequence genomic BACs from the
RPCI-23 Black 6 mouse library generated by Pieter deJong and collegues
at the Department of Cancer Genetics at Roswell Park Cancer Institute in
Buffalo, New York.
The BACPAC Resource Center has relocated to the
Children's Hospital Oakland Research Institute in Oakland, Ca., USA.

- We also are sequencing other regions of the mouse genome of high
biological interest.
- All of the RPCI-23 Female Mouse BACs that we are sequencing are from the
NHGRI Mouse Genome Project approved C57BL/6J strain, aka Black 6 and
requests for them should be directed to Dr. de Jong's BACPAC

Female 129S6/SvEvTacÊ mouse spleen genomic DNA (partially MboI
digested) was cloned into the Bam HI sites of the pPAC4 vector.
The ligation products were transformed into DH10B
electrocompetent cells (BRL Life Technologies).

Female 129S6/SvEvTac (Taconic)
mouse spleen genomic DNA was isolated and partially digested
with a combination of EcoRI and EcoRI Methylase.Ê Size
selected DNA was cloned into the pBACe3.6 vector at the EcoRI
sites.Ê The ligation products were transformed into DH10B
electrocompetent cells (BRL Life Technologies).
- RPCI - 23 Female (C57BL/6J) Mouse BAC Library
Female C57BL/6J mouse kidney and/or brain genomic DNA was isolated
and partially digested with a combination of EcoRI and EcoRI
Methylase.Ê Size selected DNA was cloned into the pBACe3.6
vector at the EcoRI sites.Ê The ligation products were
transformed into DH10B electrocompetent cells (BRL Life
Technologies).

- Recently we have begun investigating a PCR-based approach using
human chromosome 22 exon-specific primers for both PCR and custom
synthetic primer-based sequencing. Details
- We also are sequencing regions of other human and syntenic mouse chromosomes of high biological interest mapped by other collaborators.

- In accord with the Bermuda Agreement and NHGRI policy, we are depositing all our human
genomic sequence data into the
High-Throughput Genomic Sequences (HTGS) division of GenBank
as soon as a target large insert clone sequencing project can be assembled
into contigs greater than 2 kb.
