Plumbago zeylanica cDNA Sequencing
Plumbago zeylanica is a plant with dimorphic
sperm cells. Two cDNA libraries was generated from two
populations of sperm cells, Svn and Sua, in collaboration
with Xiaoping Gou in Scott Russell's
laboratory in the Department of Botany and Microbiology
at The University of Oklahoma, Norman. Random cDNA clones
are being end sequenced by
Doris Kupfer and Sunkyoung So at the Advanced Center for
Genome Technology in the Department of Chemistry and
Biochemistry at The University of Oklahoma, Norman.
Plumbago zeylanica Latest Data Release - September 2, 2003
- We now have end sequence from svn and sua cDNAs
from both ends of the directionally cloned inserts after excision into
pTriplEx2.
- Sua sperm sequences: After screening out ribosomal RNA, mitochondrial RNA, vector and small
(less than 100bp) inserts, we have 893 Phred (Phil Green, University of Washington) high quality sequences representing 324,774 total bases.
- Svn sperm sequences: After screening out ribosomal RNA, mitochondrial RNA, vector and small
(less than 100bp) inserts, we have 629 Phred (Phil Green, University of Washington) high quality sequences representing 241,007 total bases.
- All of our data is available from our ftp site, and we now have added
the ability
to perform blast searches on this data. A keyword search of a blastx search of GenBank with this data also is available but we have not yet linked this to a
unigene database as the number of EST's sequenced doesn't warrent this yet.

Obtaining the sequence data via ftp
All of the cDNA sequence data we have obtained to date is available from our
ftp site directly, by anonymous ftp to ftp.genome.ou.edu (logon as anonymous with
password your e-mail address), cd to /pub/plumbago_dir and read the 00readme
file to determine which files you should get. These files contains all
the EST's sequenced to date concatenated into a flat file either with or
without annotation, respectively.

Clarification of our naming convention
For Plumbago zeylanica
- The 3' end sequences have the notation .f1 at the end of the clone name
- The 5' end sequences have the notation .r1 at the end of the clone name
Clone Availability
At the present time, none of the cDNA clones are available for distribution.
We anticipate at some point to generate a set of arrayed cDNA clones that will be made available through the ATCC.
